Olink NPX distribution
Hi all -
Just a quick clarification regarding NPX distributions from Explore 3072. These are roughly around 0 with negative values, which differs from the distribution obtained from Target 96. These two platforms shouldn't be compared directly in general but just trying to make sense of these differences: are they due to the quantification methods (NGS vs qPCR)?

So far as I can tell, the differences in centres are only due to the normalisation performed, not to do with the physical methods at all. See https://olink.com/faq/data-normalization/ .
I have no idea why Olink have chosen to do the normalization differently for the Explore assays. I don't know how well they would compare if you subtracted the Target 96 median from the Target 96 data.
Other differences in the distribution would be due to the quantification methods, see https://olink.com/faq/can-customers-compare-and-combine-explore-runs-with-target-96-or-other-historical-target-96-studies/ , which suggests that some differences arise because the different methods have different amounts of success around the limits of detection.
